Mitochondrial reactive oxygen species modify extracellular vesicles secretion rate

چکیده
Extracellular vesicle (EV) secretion rate is stimulated by hypoxia that causes increased reactive oxygen species (ROS) production the mitochondrial electron transport chain (ETC) and hypoxia-induced factor (HIF)-1 signaling; however, their contribution to EV unknown. We found marker in our reporter cell line CD9truc-EGFP was unaffected HIF-1α stabilizer roxadustat; yet, ETC stimulation dichloroacetic acid (DCA) significantly secretion. The DCA-induced blocked antioxidant TEMPO rotenone, an inhibitor of ETC's Complex I. Under hypoxic conditions, limited reduction impedes III. To mimic this, we inhibited III with antimycin A, which ROS-dependent between I accomplished coenzyme Q created mevalonate pathway tyrosine metabolites. Blocking early step using pitavastatin augmented secretion, 4-nitrobenzoate—an condensation metabolites—increased Our findings indicate hypoxia-mimetics targeting modify ROS produced a potent stimulus for

متن کاملMitochondrial metabolism of reactive oxygen species.
For a long time mitochondria have mainly been considered for their role in the aerobic energy production in eukaryotic cells, being the sites of the oxidative phosphorylation, which couples the electron transfer from respiratory substrates to oxygen with the ATP synthesis. Subsequently, it was showed that electron transfer along mitochondrial respiratory chain also leads to the formation of rad...
متن کاملMitochondrial reactive oxygen species and Ca2+ signaling.
Mitochondria are an important source of reactive oxygen species (ROS) formed as a side product of oxidative phosphorylation. The main sites of oxidant production are complex I and complex III, where electrons flowing from reduced substrates are occasionally transferred to oxygen to form superoxide anion and derived products. These highly reactive compounds have a well-known role in pathological...

Mitochondria produce reactive oxygen species (mROS) as a natural by-product of electron transport chain activity. While initial studies focused on the damaging effects of reactive oxygen species, a recent paradigm shift has shown that mROS can act as signaling molecules to activate pro-growth responses. Cancer cells have long been observed to have increased production of ROS relative to normal ...

The inner membrane of a plant mitochondrion contains the mitochondrial electron transport chain (mtETC), consisting of protein complexes that use an energy source-derived reductant to form a proton gradient across the membrane. This proton gradient drives ATP synthesis, a primary mitochondrial function. Ultimately, electrons in the mtETC are transferred through one of two pathways from ubiquino...
